Relative firefly luciferase activities in Arabidopsis protoplasts transfected with various pDuEx vectors in 96-well plates were compared. A cDNA encoding firefly luciferase was inserted in the pDuEx-Prey vector (pDuExAn6GL3) and pDuEx-Bait vector (pDuExDc6GL3) respectively.? As an independent gene, a cDNA encoding an Arabidopsis transcription factor, PERIANTHIA (PAN, At1g68640), was inserted in the bait (pDuExDc6PAN) and prey (pDuExAn6PAN) vectors respectively. The prey and bait vectors carrying firefly luciferase were recombined with bait and prey vectors carrying the transcription factor respectively (pDuExAn6GL3-Dc6PAN and pDuExAn6PAN-Dc6GL3). ?Five thousands of Arabidopsis protoplasts were transfected with 5 mg of each vectors in 96-well plates and the luciferase activities were measured 16 hours after the infections (sample number, n = 20).? In mock samples, no DNA vector was added during the transfection. Luminescence intensities are expressed as % of intensity in the protoplasts transfected with pDuExAn6GL3. The size of each DNA vector is shown under the column with parentheses.
